体内研究 DuP 734可有效阻断甲磺酸卡林诱导的抓挠(ED50=0.35 mg/kg,p.o.)和攻击性活动(ED50=1.9 mg/kg,p.o.),作为阿扑吗啡拮抗剂(ED50=12 mg/kg,p.o.)[1]。给予dup734可显著拮抗[3H]dup734和[3H](+)-SKF 10047与脑西格玛受体的结合,其ID50值分别为0.02和0.07mg/kg(0.07和0.25μmol/kg)[2]。静脉给药后,DuP 734在小鼠、大鼠、比格犬和食蟹猴体内的分布特点是全身血浆清除率高(46至87 mL/min/kg)和较大的稳态分布体积(3.6至6.8 L/kg)。最终消除半衰期为50-83min。小鼠和大鼠胃肠道吸收水溶液的速度非常快,在给药后5 min和20 min内达到峰值DuP 734血浆浓度。狗和猴子的血浆浓度峰值分别在45分钟和130分钟内达到。在3.1至30.1mg/kg剂量下,小鼠的绝对生物利用度为29%至46%。dup734在大鼠和狗体内的生物利用度分别从12.5~50mg/kg和1~3mg/kg增加到10%和14~72%。在9.3mg/kg的dup734剂量下,猴子的生物利用度为30.5%。DuP 734的剂量依赖性药代动力学在所有研究动物物种的窄剂量范围内观察到[3]。
