体外研究	β-淀粉样蛋白(1-40)和(1-42)是老年斑淀粉样蛋白的主要成分,是存在于脑中的生理肽,脑脊液(CSF)和血浆。 CSFβ-淀粉样蛋白(1-40)和(1-42)的水平在正常衰老中显示出U形自然过程[1]。在大鼠脑室中长期输注β-淀粉样蛋白(1-40)14天降低海马中可溶性蛋白激酶C(PKC)的活性。在用0.5微摩尔佛波醇-二丁酸酯(PDBu)的急性刺激下,用β-淀粉样蛋白(1-40)处理的大鼠的海马切片中PKC亚细胞易位至膜组分被完全消除[2]。
体内研究	慢性输注β-淀粉样蛋白(1-40)进入大鼠脑室导致空间和非空间记忆形成的缺陷[2]。 β-淀粉样蛋白(1-40)的慢性治疗不会显着改变杠杆按压性能,但是在慢性每日方案终止后30天,性能显着下降。注射到背海马中的可溶性非聚集形式的β-淀粉样蛋白(1-40)似乎对大鼠的表现或短期工作记忆没有行为影响,但多次重复注射在几周后产生性能下降。通过留置套管重复注射β-淀粉样蛋白(1-40)显示出阿尔茨海默病动物模型的发展前景[3]。
动物实验	大鼠：HPLC缓冲液确保β-淀粉样蛋白（1-40）在注射前不会在溶液中聚集，β-淀粉样蛋白（1-40）和载体在实验期前20分钟双侧输注到海马体内，体积为1 ，每侧2和3μL，速率<1μL/ min。使用不同体积的1μM溶液。体积（剂量）以随机顺序给出，并且在β-淀粉样蛋白（1-40）或载体注射之间插入至少三次假注射疗程。在急性注射方案下，所有大鼠均接受所有剂量的β-淀粉样蛋白（1-40）[3]。
