12S-HHT用途

12S-HHT(12(S)-HHTrE)是前列腺素H2(PGH2)的酶促产物,来源于环氧化酶(COX)介导的花生四烯酸代谢。12S-HHT是BLT2的内源性配体,在体内完全激活BLT2。12S-HHT抑制紫外线诱导的角质形成细胞中IL-6的合成,发挥抗炎活性[1][2]。

[体外研究]

12S-HHT(0-150nm；3小时)通过减弱UVB诱导的HaCaT细胞中IL-6合成而具有抗炎活性[2]。12S-HHT通过上调MKP-1抑制UVB刺激的p38 MAPK/NF-κB通路,从而抑制IL-6合成[2]。12S-HHT是BLT2的内源性激动剂[3]。Western Blot分析[2]细胞系：HaCaT细胞浓度：0,12.5,25,75或150 nM培养时间：3小时结果：UVB(5 mJ/cm2)照射显著上调IL-6的合成和释放,12-HHT以浓度依赖性方式抑制IL-6的合成和释放。
