K+ channel[1]

体外研究 Almitrine通过降低其开放概率抑制高电导(152±13 pS),Ca2 +依赖性K +通道的活性。效果的IC50值为0.22μM。在GH3细胞中也观察到Almitrine对Ca2 +依赖性K +通道的抑制作用。浓度高达10μM的Almitrine不会影响大鼠或兔细胞中的全细胞电压依赖性K +,Ca2 +或Na +电流。然而,这种浓度的Almitrine显着抑制大鼠化学感受器细胞中K +电流的Ca2 +依赖性成分[1]。
体内研究 Almitrine通过外周动脉化学感受器起作用,提高颈动脉窦神经输出和每分钟通气量。 Almitrine还具有肺血管作用,引起剂量依赖性收缩和扩张。在低剂量时,Almitrine可增强缺氧性肺血管收缩,并可改善整体通气/灌注比[2]。
动物实验 使用大鼠[2] 8周龄雄性spf Wistar大鼠。用硫代巴比妥酸盐肌动蛋白(BYK,100mg / kg,ip)麻醉大鼠。检测通气反应与缺氧和间歇性(2分钟,1分钟休息)低剂量(10μg/ kg每分钟)和高剂量(80μg/ kg每分钟)输注S9581或Almitrine的相互作用在对照和慢性缺氧大鼠中。静脉输注S9581或Almitrine(100μg/ mL)。通过使氧气 - 氮气混合物以3-41分钟-1的流速通过气管端口来控制灵敏的氧气水平[2]。
