[体外研究]

ACTH(1-17)是hMC1R的有效激动剂。 ACTH(1-17)显示对hMC1R的高亲和力,Ki值为0.21±0.03 nM,略高于α-MSH [1]的0.13±0.005 nM。 ACTH(1-17)即使在大鼠垂体培养中使用微摩尔浓度的肽时也会诱导生长激素分泌的轻微且不显着的增加[2]。

[体内研究]

当夜间动物变得活跃时,在每日黑暗跨度开始后2小时施用ACTH(1-17)时,注意到DNA标记的抑制。当在该昼夜节律阶段施用时,特别是较大剂量与持续24小时的DNA标记的抑制相关。当4小时前注射相同剂量时,抑制作用要短得多[3]。

[细胞实验]

在不同浓度的ACTH（1-17）（0.1nM-1μM）存在下孵育大鼠垂体细胞。每种浓度都记录了生长激素分泌的显着增加[1]。

[动物实验]

小鼠[3]在时间药理学给药方案中测试ACTH（1-17）对CD2F1小鼠干骺端骨中DNA标记率的影响。已经调整为12小时光照/ 12小时黑暗时间表的小鼠组在六个不同时间点之一注射，间隔4小时，一小时24小时跨度低（0.021IU / kg）或高剂量（20 IU / kg）ACTH（1-17）。对照组在相应的时间点接受安慰剂注射。亚组小鼠注射[3H]胸苷（[3H] Tdr），以跟踪ACTH后1分钟和2小时，4小时，8小时，12小时和24小时胫骨近端干骺端DNA标记的变化（1-17）或安慰剂治疗[3]。
