[体外研究]

VAS2870有效抑制PDGF-BB依赖的NADPH氧化酶活化和随后的细胞内ROS产生。此外,VAS2870抑制PDGF-BB依赖性趋化性,但不抑制DNA合成。与VAS2870(10和20μM)预孵育完全消除了PDGF介导的NADPH氧化酶活化和ROS产生。与VAS2870(0.1-20μM)预孵育不影响PDGF诱导的细胞周期进展。然而,它以浓度依赖性方式消除了PDGF依赖的VSMC趋化性(在10μM时100％抑制)[1]。 VAS2870抑制FaO大鼠肝癌细胞中细胞数量的剂量依赖性自分泌增加,并且当以25mM使用时几乎完全阻断ROS产生和胸苷掺入。 VAS2870阻断FaO大鼠肝癌细胞的血清依赖性细胞生长。 VAS2870抑制不同人肝细胞癌(HCC)细胞系的增殖。 VAS2870预处理可增强TGF-β介导的FaO大鼠肝癌细胞凋亡[2]。

[激酶实验]

在含有1mM EGTA，蛋白酶抑制剂，150mM蔗糖，5μM光泽精和250μMNADPH作为底物的50mM磷酸盐缓冲液（缓冲液A），pH 7.0中，通过光泽精增强化学发光测量NADPH氧化酶活性。通过血清剥夺使静止细胞饥饿24小时并如所示进行处理，用冰冷的磷酸盐缓冲盐水（PBS）（pH 7.4）洗涤两次，并收获。低旋转离心后，将沉淀重悬于冰冷的缓冲液A中，缺乏光泽精和底物。然后，裂解细胞并使用Bradford测定法测定总蛋白质浓度并调节至1mg / mL。在闪烁计数器中使用NADPH（100μM）作为底物，在6分钟内一式两份测量100μL等份的蛋白质样品。每隔2分钟收集数据以测量NADPH氧化酶活性的相对变化[1]。

[细胞实验]

为了测试自分泌生长，将细胞在40％汇合时剥夺血清，并且当指示时，在血清剥夺前30分钟加入NADPH氧化酶抑制剂Apocynin（300mM）或VAS2870并沿实验保持。对于TGF-b实验，在存在或不存在EGFR抑制剂AG1478（20mM）或VAS2870（25mM）的情况下，将70％汇合的细胞剥夺血清16小时并用2ng / mL TGF-β处理。在TGF-β之前30分钟加入[2]。
