[体外研究]

克洛替诺(10μM)显著抑制hERG表达细胞中的电压门控外向电流[1]。克洛替诺(0-1000μM)抑制IC50值为1.9μM的hERG转染细胞的外向电流[1]。

[体内研究]

克洛替诺(1-20 mg/kg;静脉注射一次)对豚鼠的气道没有明显影响[2]。克洛替诺(1和10 mg/kg;静脉注射一次)影响豚鼠的心率和平均血压[3]。动物模型:麻醉豚鼠[2]剂量:1-20mg/kg给药:静脉注射;结果:对气道无明显影响,但高剂量对心血管系统有显著影响。动物模型:麻醉闭胸豚鼠[3]剂量:1和10 mg/kg给药:静脉注射;结果:1 mg/kg剂量可降低心率,10 mg/kg剂量可以降低心率和平均血压。
