[体外研究]

Batimastat(BB-94)是一种有效的基质金属蛋白酶抑制剂,具有意想不到的结合方式。 Batimastat抑制明胶酶A和B,IC50值分别为4 nM和10 nM。具有结构相似的胶原酶Ht-d的IC50为6nM,其与MMP-1(3nM),MMP-8(10nM)和MMP-3(20nM)的值相当[2]。从血浆酸性金属蛋白酶抑制剂Batimastat(BB-94,IC50 = 230 nM)可有效阻断从细胞系Karpas299脱落的CD30 [3]。

[体内研究]

腹腔注射Batimastat(BB-94)可有效阻断人卵巢癌异种移植物和小鼠黑色素瘤转移的生长,延迟原发性肿瘤在人乳腺癌原位模型中的生长,无细胞毒性,且不影响mRNA水平[2]。 Batimastat(BB-94)是一种合成的基质金属蛋白酶抑制剂,在各种肿瘤模型中显示出抗肿瘤和抗血管生成活性。用顺铂(每隔一天口服60mg/kg,总共8次注射)同时用顺铂(4mg/kg静脉注射,每7天注射3次)完全阻止两种异种移植物的生长和扩散,以及所有动物在第200天都活着并且健康[4]。 Kaplan-Meier生存分析(48小时)显示,与对照组(75％)相比,用Batimastat(BB-94)治疗的动物存活率增加(95.2％),差异几乎具有统计学意义(p = 0.064)[ 5]。在E2施用后4小时,在盐水或Batimastat(40mg/kg)预处理的动物中分析基质密度,在激素处理后观察到胶原密度处于最低的时间点[6]。

[动物实验]

小鼠[5]使用6周龄雌性BALB / c小鼠。在感染前和感染后24小时，用Batimastat（BB-94,50mg / kg）腹膜内处理小鼠。将Batimastat在DMSO中悬浮于50mg / mL并在-20℃下冷冻保存。在使用之前，将其在磷酸盐缓冲盐水（PBS）中稀释20倍，并将500μL注射到动物中。对照小鼠注射500μL含5％DMSO的PBS。在ic攻击后48小时处死动物。大鼠[6]雌性Sprague-Dawley大鼠在尸检时在指定的时间间隔内通过腹膜内（ip）注射给予单一生理剂量的E2（40μg/ kg，在0.9％NaCl，0.4％EtOH载体中）。已显示E2的这种体内剂量水平诱导子宫湿重，组织结构和雌激素受体激活特征的基因表达的变化。对于所有其他实验，在组织收获前4小时腹膜内施用单个40μg/ kg推注的E2，而对照动物仅在所有研究中接受载体。 Batimastat以剂量水平ip（40mg / kg，在1×PBS，0.1％Tween-20载体中）施用，显示在E2或盐水对照之前4小时有效抑制体内MMP。
