作为重要的第二信使，cGMP是细胞外信号（例如一氧化氮和利尿钠肽）的主要细胞内介质。环状GMP可与三种类型的细胞内受体蛋白相互作用：cGMP依赖性蛋白激酶、cGMP调节的通道、以及cGMP调节的环核苷酸磷酸二酯酶。cGMP水平在体内升高的主要作用是刺激cGMP依赖性蛋白激酶（PKG）。
