体外研究 ARN5187三盐酸盐(化合物1)(0-100µM;48 h)对BT-474细胞显示细胞毒性,EC50为23.5µM,对BT-474和HMEC细胞显示IC50为30.14µM,>100µM[1][2]。ARN5187三盐酸盐(0-100µM)在HEK-293细胞中以浓度依赖性方式激活RevRE reporter[1]。ARN5187三盐酸盐(25,50µM)是一种溶酶体非依赖性REV-ERB拮抗活性[1]。ARN5187三盐酸盐(50µM;24 h)显示出自噬抑制[1]。ARN5187三盐酸盐(50µM;2、8、24小时)影响自噬形成和成熟[1]。细胞毒性试验【1】细胞系:BT-474细胞浓度:0-100µM孵育时间:48小时结果:EC50为23.5µM。Western Blot分析【1】细胞系:BT-474细胞浓度:50µM孵育时间:24小时结果:显著增加α-LC3-II、α-p62、α-裂解PARP的表达。RT-PCR【1】细胞系:BT-474细胞浓度:25,50µM孵育时间:结果:BMAL1、PER1和PEPCK的表达呈剂量依赖性显著增强。
