1)2,6-二氯喹喔啉的制备 4-氯-2-硝基苯胺在苯或甲苯溶剂中、催化剂吡啶存在下与双乙烯酮反应,生成相应的α-乙酰物。

2)上述产物经碱性水溶液处理,生成相应的喹喔啉酮氮氧化物。以甲苯-水为溶剂,在氢氧化钠(或氢氧化钾)存在下,于60~70℃反应1h制得。

3)氮氧化物在碱水溶液中,于室温下,与一定量的30%硼氢化钠反应,得6-氯-2(1H)-喹喔啉酮。

4)6-氯-2(1H)-喹喔啉酮在DMF溶液中与氯化亚砜作用,得2,6-二氯喹喔啉。

5)喹禾灵的制备 在氮气保护下、DMF中,2,6-二氯喹喔啉与对苯二酚缩合(碱性条件)可得到4-(6-氯-2-喹喔啉氧基)酚,且收率较高。

6)然后在乙腈溶液中,以碳酸钾为缚酸剂,4-(6-氯-2-喹喔啉氧基)酚与α-卤代丙酸乙酯缩合,合成喹禾灵。

7)喹禾灵也可用2,6-二氯喹喔啉与α-对羟基苯氧基丙酸乙酯在碳酸钾存在下,于乙腈溶液中回流24h直接作用制得。其中α-对羟基苯氧基丙酸乙酯可通过下列途径制得。
