[体外研究]

将8μMGSH样品溶液分别用8,80,200,400,800,1600,2000,2500μM5-IAF衍生化,并用CE-LIF分析。使用400和800μM5-IAF获得最高信号强度[2]。

[激酶实验]

将HepG2细胞在1mL 2％乙腈水溶液中裂解，并转移至1.5mL离心管中。使用另外的200μL2％乙腈冲洗培养皿并与细胞提取物组合。为了测量游离GSH浓度，在快速混合后立即将三份80μL细胞提取物转移至0.5mL离心管中。然后加入20μL50μMNAC，然后向混合物中加入100μL400μM5-IAF。衍生化在室温下在黑暗中进行1小时，之后将混合物以9000×g离心5分钟以沉淀不溶性蛋白质。将10μL上清液用水稀释100倍并立即用CE-LIF分析[2]。
