C2 Ceramide (Ceramide 2) 是一种角质层的主要脂质,也是一种蛋白质磷酸酶 1 (PP1) 的激活剂。C2 Ceramide 还可激活 PP2A 和神经酰胺激活的蛋白磷酸酶 (CAPP)。C2 Ceramide 可诱导细胞分化和凋亡 (apoptosis),抑制线粒体呼吸链复合物 III (mitochondrial respiratory chain complex III)。C2 Ceramide 是一种皮肤调理剂,可保护表皮屏障免受水分流失。

[体外研究]

C2神经酰胺(5nm-200μM；24小时；原代小鼠成骨细胞)治疗(≤500nm)促进成骨细胞活性,而浓度≥2μM以剂量和时间依赖的方式显著降低成骨细胞活性[1]。在成骨细胞中,C2神经酰胺浓度为50μM和100μM时,C2神经酰胺分别使细胞质组蛋白相关DNA片段增加5.7倍和11.2倍。在这些高浓度下,C2神经酰胺是成骨细胞凋亡的有效诱导剂[1]。C2神经酰胺上调人牙髓细胞(hdpc)血管生成基因mRNA的表达,增加内皮细胞的迁移和毛细血管的形成,而PP1小干扰RNA则相反。人牙髓细胞(HDPCs)增加骨形态发生蛋白2、Smad磷酸化1/5/8、runt相关转录因子2和osterix的mRNA表达[2]。细胞活力测定[1]细胞系：原代小鼠成骨细胞浓度：5n M-200μM培养时间：24小时结果：小鼠成骨细胞暴露于低浓度的5-500 n M时,其存活率呈剂量依赖性增加。20-200μM浓度增加导致线粒体呈剂量依赖性减少琥珀酸脱氢酶活性与成骨细胞存活。

[体内研究]

PP1激活剂C2神经酰胺提高碱性磷酸酶活性,矿化结节形成,牙本质基质蛋白1和牙本质涎磷蛋白mRNA表达。相反,PP1小干扰RNA的敲除抑制成牙本质细胞分化[2]。
