[体外研究]

荧光素染料是常见的荧光探针。它在氩激光波长(488nm)下具有非常高的摩尔吸光系数,大的荧光量子产率和高光稳定性使其成为非常有用和灵敏的荧光标记。荧光素可以许多衍生物商购获得,例如异硫氰酸荧光素和荧光素琥珀酰亚胺酯,它们可以共价连接到大分子和氨基酸上。可以以非常高的灵敏度检测标记的分子,其用于例如毛细管电泳。荧光素的发射光谱广泛地重叠四甲基罗丹明的吸收光谱,四甲基罗丹明是一种相关的强荧光染料,使得该对非常适合于能量转移实验以确定标记的大分子内和之间的距离。水溶液中的荧光素以阳离子,中性,阴离子和双阴离子形式存在,使其吸收和荧光性质强烈依赖于pH [1]。

荧光素制备

1.将间苯二酚加热至150℃,使之全部熔融,边搅拌边加入理论量的邻苯二甲 酸 酐,混匀并熔融后 升温至185℃,保温半小时,然后慢慢加入适量新焙烧的无水氯化锌,当完全溶解后,逐渐升温至210~215℃,整个过程均需不停地搅拌:

当反应液开始变稠时,停止搅拌,继续在此温度下加热至完全固化,研碎后得粉状粗品。将粉状粗品与稀盐酸混合加热煮沸,以浸出氯化锌和剩余的间苯二酚,抽滤后水洗、干燥,再用乙醇提取,晾干后即得红色固体荧光黄。
