[体外研究]

PACAP(1-38),人,绵羊,大鼠是垂体腺苷酸环化酶激活多肽的片段[1]。 PACAP(1-38)显示来自包括内分泌胰腺在内的各种组织的膜中PACAP特异性(PAC1)受体的高亲和力[2]。在体外,PACAP(1-38)使由组胺和乙酰胆碱预收缩的豚鼠和兔气管平滑肌松弛。 PACAP(1-38)也增加气管平滑肌中腺苷3'：5'-环一磷酸(环AMP),为PACAP(1-38)的松弛作用提供了可能的机制[3]。

[体内研究]

假手术中单独的PACAP(1-38)不会导致任何视网膜层的变化。 PACAP(1-38)溶于溶液眼科和苯扎克诺,可在双侧颈总动脉闭塞(BCCAO)视网膜中对视网膜产生显着保护作用;与对照视网膜相比,用PACAP(1-38)滴眼液治疗的视网膜具有保留的结构。与假对照相比,BCCAO视网膜中OLM-ILM(外界膜-内界膜)距离减少49.7％(p <0.001),但用PACAP治疗的眼睛仅为40.6％(p <0.001)(1) -38)滴眼液。内核层(INL)(BCCAO：38.5％,PACAP(1-38)：30.5％; p <0.001)和内丛状层(IPL)(BCCAO：64.8％, PACAP(1-38)：38.2％; p <0.05),而在外核层(ONL)中未观察到统计学上显着的损害衰减(BCCAO：36.5％,PACAP(1-38)：37.7％)或外丛状层(OPL)(BCCAO：53.0％,PACAP(1-38)：48.2％)。神经节细胞层(GCL)中的细胞数量在BCCAO后显着降低52.4％(p <0.05),并且通过PACAP(1-38)滴眼液显着改善(降低25.9％; p <0.05)[4] ]。

[动物实验]

在12/12小时的光/暗循环下喂食大鼠[4] Wistar大鼠（n = 20：n = 12用于组织学分析，n = 8用于免疫组织化学分析），体重250-300，并随意浇水。手术后1分钟内，右眼用PACAP（1-38）滴眼液（1μg/滴）治疗。使用的载体是浓度为0.005％的苯扎氯铵，因为它是用PACAP1-27滴眼液实现神经保护的最有效载体。左眼用作对照，仅用车辆治疗。一组动物用作假手术组，其经历麻醉和外科手术的所有步骤，除了颈动脉结扎。大鼠每天治疗两次，连续5天[4]。
