[体外研究]

残基7后的胰高血糖素样肽的序列显示与胰高血糖素和分泌素肽家族的其他生物活性成员,特别是葡萄糖依赖性促胰岛素肽(GIP)的相似性。该序列保存得特别好,在非常原始的鳗鱼的前胰高血糖素中与GLP-1显示66％的核苷酸同源性。这种7-36的GLP-1序列在体外是一种有效的胰岛素释放肽[1]。胰高血糖素样肽(GLP)I(7-36),酰胺是胰高血糖素前体的组织特异性翻译后加工的产物。它从肠内分泌L细胞释放,并刺激胰岛素分泌。 DPP IV是人血清中GLP-1(7-36)酰胺的主要降解酶。二肽基肽酶IV可以启动人血清中GIP和GLP-1(7-36)酰胺的代谢[2]。

[体内研究]

胰高血糖素样肽(GLP)I(7-36),酰胺是一种生理性肠促胰岛素激素,在从肠道下部营养摄入后释放,并在升高的血浆葡萄糖浓度下刺激胰岛素分泌。外源性GLP-1(7-36酰胺)是在控制不良的2型糖尿病受试者中使空腹血浆葡萄糖浓度正常化的有效方法[3]。外源性施用GLP-1-(7-36)酰胺在体内极其不稳定,在sc或iv施用后超过80％被切割成GLP-1-(9-36)酰胺[4]。

[细胞实验]

为了测试灌注培养基本身是否存在GLP-1降解活性，合成的胰高血糖素样肽（GLP）I（7-36），酰胺在体外与从中收集的培养基一起孵育（在37℃下30分钟）。动脉线（即在通过组织之前）和静脉线，并进行HPLC和RIA分析[4]。
