[体外研究]

CA-074是E-64的合成类似物,E-64是一种天然的肽基环氧化物,不可逆地抑制大多数已知的溶酶体半胱氨酸蛋白酶,并且是通过合理的药物设计开发的,利用组织蛋白酶B的二肽基羧肽酶活性.CA-074可用于选择性抑制活细胞内的组织蛋白酶B,只要实验条件允许药物显着的液相内吞作用[2]。 CA-074以2至5nM的Ki抑制组织蛋白酶B,而组织蛋白酶H和L的初始Kis为约40-200μM。 CA-074对纯化的大鼠组织蛋白酶B的抑制作用比对组织蛋白酶H和L的抑制作用高10000-30000倍[1]。

[体内研究]

向大鼠腹腔注射化合物CA-074有效地和选择性地抑制组织蛋白酶B的活性[1]。在缺血性损伤后立即静脉内给予CA-074,在第8天,在经历20分钟脑缺血的8只猴子中,67％的CA1神经元免于延迟神经元死亡：八只猴子中的三只中的抑制程度非常好,八只猴子中的五只中有效良好[3]。

[动物实验]

大鼠：将化合物CA-074或CA-030或E-64腹膜内注射为含有DMSO的盐水溶液。注射剂量为8mg / 100g体重。注射后6小时，通过向头部吹气杀死大鼠，并用盐水灌注肝脏，取出，称重并在冰上冷却。将4g肝脏的样品切碎并均质化。将匀浆以800×g离心15分钟，并将上清液以12000×g离心30分钟。将沉淀物悬浮于2mL 0.05M乙酸盐缓冲液（pH5.0）中，并冻融以测量组织蛋白酶B，H和L活性[1]。
