[体外研究]

Phenylbiguanide(1-苯基双胍)是一种5-HT3受体选择性激动剂,r5-HT3A(b),h5-HT3A,m5-HT3A(b)的pEC50分别为5.57,4.07和4.47 [1]。研究了苯基双胍(1-苯基双胍盐酸盐)促进HT29细胞生长的作用。培养48小时后,苯基双胍引起HT29细胞的剂量依赖性增殖。最大增殖在5HT浓度12.5μM(P≤0.01)。 Phenylbiguanide在浓度为3.125μM(P≤0.05)和6.25μM(P≤0.01)时显着刺激细胞生长[2]。

[体内研究]

在麻醉的小鼠中,已知刺激心肺传入C-纤维的药物Phenylbiguanide(PBG)注入心脏的右心房,并在中枢神经系统的特定区域内定位c-Fos表达。心房内注射PBG产生反射性心肺功能反应,包括明显的心动过缓和呼吸抑制[3]。

[细胞实验]

用磷酸盐缓冲盐水（PBS）洗涤HT29细胞，并用0.5％胰蛋白酶溶液以50-60％汇合度收获。然后将细胞以104细胞/孔的密度在96孔板中加入孔中至终体积为100μL/孔。在37℃，5％CO 2气氛中培养24小时后，用含有5HT盐酸盐，盐酸苯基双胍的200μL新鲜培养基替换培养基，浓度为：3.125,6.25,12.5,25,50和100μM。。仅在培养基中培养的细胞用作阴性对照。在37℃，5％CO 2气氛中培养48小时后，除去培养基，将8μLMTT试剂（在PBS中稀释，浓度为4mg / mL）加入到50μL新鲜培养基中。终浓度为0.55 mg / mL。最佳潜伏期是在试验研究中确定的[2]。

[动物实验]

小鼠[3]在整个研究中使用成年雄性指定的无病原体（SPF）BALB / c小鼠（28-33g; n = 10）。所有小鼠都可以在光线下（12：12小时光照/黑暗循环，在0700小时开灯）和温控（21-23°C）环境中自由获取水和食物。将小鼠随机分配到两个治疗组，1）心房内注射0.9％盐水（对照，n = 5）或2）心房内注射含有苯基双胍（n = 5）的载体。对于PBG注射组，给小鼠注射有效剂量的苯基双胍（1-1.5μg，在10-15μL盐水中）。这总共重复五次，每次注射间隔8-10分钟。对于对照小鼠，每只小鼠接受5次盐水注射（每次10-15μL），也间隔8-10分钟[3]。
