Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs). Caspase activity assay[1]: 1. Incubate the cells according to your normal protocol. 2. Wash cells once with PBS and centrifugate. 3. Resuspend cells in PBS at a concentration of 1× 107 cells/mL. 4. Prepare the standard reaction buffer which includ 100 μM caspase-8 substrate AC-LETD-AFC. Prepare correspounding standard reaction buffer according to your protocol. 5. Add 15 ul of the cells suspension to a microplate, and mixed with the appropriate peptide substrate dissolved in a standard reaction buffer. 6. Measure substrate cleavage with a microplate reader with excitation wavelength of 360 nm and emission at 460 nm.
