体外研究	治疗72小时后,TPC-1细胞对RPI-1的生长抑制作用敏感,IC50为5.1μM。RPI-1(7.5-60µM)抑制TPC-1细胞中的Ret/Ptc1自磷酸化。TPC-1细胞培养条件下的RPI-1抑制效应导致JNK2和AKT相关通路的抑制[1]。在表达Ret突变体的NIH3T3细胞中,细胞增殖的RPI-1 IC50值为3.6µM,而在未转染的NIH3T3细胞中,RPI-1 IC50值为16µM；在软琼脂中,表达Ret突变体的NIH3T3细胞和转染H-RAS的NIH3T3细胞中,集落形成的RPI-1 IC50值分别为2.4µM和26µM。在表达Ret突变体的NIH3T3细胞中,RPI-1处理24小时后,检测不到Ret蛋白和酪氨酸磷酸化。在TT细胞中,RPI-1抑制增殖、Ret酪氨酸磷酸化、Ret蛋白表达以及PLCgamma、ERKs和AKT的激活[2]。
体内研究	RPI-1(50,100 mg/kg；p、 o。；每天两次,持续10天)抑制TT异种移植物的肿瘤生长81%[2]。动物模型：8-11周龄雌性裸鼠CD-1(含TT细胞)[2]剂量：50100mg/kg给药：P.o。；每天两次,持续10天结果：观察到对肿瘤生长的剂量依赖性影响,治疗10天后,250 mg/kg/天的剂量比2100 mg/kg/天的剂量对肿瘤重量的抑制作用更小。
