阿苯达唑制备

方法1:硫化钠溶于95%乙醇中,在50℃下加入2-硝基-4-硫氰酸酯基苯胺。在60℃下搅拌,冷却到40℃,一次性加入溴丙烷。于40℃反应后再升至60℃反应。加入硫化钠,在80℃下反应。减压蒸出乙醇,加入水。冷却,用氯仿提取。提取液水洗,干燥,加入S-甲基-N,N-二(甲氧羰基)异硫脲和冰醋酸,回流。蒸出氯仿,加入甲醇。冷却,抽滤,用甲醇洗涤,干燥,得阿苯达唑粗品。


方法2:在搅拌和冰浴冷却下,往氯磺酸中缓慢加入多菌灵,保持内温15~20℃。加毕在40℃下搅拌。在0℃以下,把反应液缓慢滴到95%乙醇中。滤集沉淀,以少量乙醇洗,干燥,得氯磺化产物,收率90。8%。把该氯磺化产物加到甲酸、水和40%氢溴酸的溶液中,再加金属铝粉。在35~40℃下搅拌,再在60~65℃下反应。冷却后过滤,滤液用40%氢氧化钠调至Ph值3。滤集沉淀,洗涤,干燥,得巯基化物,收率82.5%。将该巯基化物加到氢氧化钠水溶液中,活性炭脱色。加入溴化四丁基铵,再加入溴丙烷的甲醇溶液,在35~40℃下反应。冷却,滤集沉淀,洗至中性,干燥,得白色结晶的阿苯达唑,收率87.5%,熔点207~209℃。


多菌灵在冰乙酸存在下,和硫氰酸钠反应,引入硫氰基后,再用硫化钠还原后和溴丙烷反应,也得阿苯达唑。
阿苯达唑可用下列方法精制:粗阿苯达唑(以干品计)和85%工业甲酸(1:3.5)一起搅拌升至60℃,待粗品完全溶解后,依次加入连二硫酸钠、活性炭和EDTA。继续搅拌升温至80℃保温。抽滤,滤液倾入去离子水中,在搅拌下用18%~20%氨水中和至Ph值6.5。抽滤,干燥,得白色产品,熔点206~212℃,精制收率93%~94%。
