[体外研究]

SU9516显示针对PKC,p38,PDGFr和EGFR的轻微活性,IC 50> 10,> 10,18和>100μM。 SU9516(5μM)降低pRB的cdk2特异性磷酸化并抑制RKO细胞中的细胞周期进展。 SU9516(5μM)也诱导RKO和SW480细胞凋亡[1]。 SU9516(5μM)导致HT-29细胞中pRb/E2F复合物形成增强。 SU9516增强多蛋白复合物中E2F物种的存在[2]。 SU9516(5μM)快速诱导细胞色素释放,Bax线粒体易位和凋亡,同时显着下调抗凋亡蛋白Mcl-1。 SU9516引起人白血病细胞中Mcl-1 mRNA水平的下调。此外,SU9516处理导致活性氧物质产量显着增加[3]。

[激酶实验]

激酶测定在96孔聚丙烯板中进行。每个反应含有2μg组蛋白H1，终浓度为10μM[-33P] ATP（0.2μCi/孔），10 mM MgCl2,1mM DTT，0.01％Triton X-100和10％甘油，40μL体积。加入20μL酶（6ng cdk2 /孔，最终浓度为1.6nM）开始反应，该酶先前在相同缓冲液中以1：50-1：200稀释，并允许进行1小时。在室温下。通过添加0.01mL 10％磷酸终止反应，并将25μL反应混合物转移至P30磷酸纤维素滤垫纸中。将过滤垫用1.0％磷酸洗涤三次，空气干燥，然后在液体闪烁计数器中计数放射性。

[细胞实验]

将RKO细胞和SW480细胞以1×10 4个细胞/孔接种在96孔板中的重复（n = 6）中，并使其附着过夜。将SU9516以0.05μM至50.00μM的浓度添加24小时，然后用PBS洗涤细胞两次，并用完全培养基补充细胞。在药物去除后0,4和7天固定细胞，并使用改良的SRB细胞毒性测定法测定蛋白质水平。将细胞在10％三氯乙酸中固定1小时，在蒸馏水中洗涤，并在0.4％SRB /乙酸中染色30分钟。然后将细胞在0.1％乙酸中洗涤，溶解于10mM Tris（pH 9）中，并在Bio-Rad 360酶标仪上在595nm处分析。所有实验重复至少三次。
