[体外研究]

苯丁酸氮芥可通过碱化诱导的交叉相互作用剥夺DNA分子互补链的功能,然后抑制肿瘤细胞增殖。苯丁酸氮芥(0,2.5,5,10μM)对Raji细胞凋亡具有轻微抑制作用,但有效增加Raji细胞中DR4和DR5 mRNA的表达。苯丁酸氮芥(10μM)与肿瘤坏死因子(TNF)相关的凋亡诱导配体(TRAIL,80 ng/ml)对Raji细胞凋亡和抑制增殖具有协同作用[1]。

[体内研究]

苯丁酸氮芥(0.2 mg/kg,po)与左旋咪唑(5 mg/kg)联合使用可增强艾氏腹水癌的抗癌作用,从而提高Ehrlich腹水癌的凋亡率和小鼠的存活率。然而,苯丁酸氮芥对小鼠的肝脏和肾脏有不良影响[2]。

[细胞实验]

将对数生长期的培养细胞用胰蛋白酶消化成单细胞悬浮液,并以每孔1000个密度接种到96孔板中。将板置于具有5%CO 2的37℃室中。在附着生长24小时后,用0,20,40和80ng / mL的TRAIL或0,2.5,5和10μM的苯丁酸氮芥处理细胞48小时。向每个孔中加入10μLCCK-8试剂,然后在37℃下孵育4小时。然后通过微板读数器测量450nm处的吸光度值。在每个处理组中进行六个平行样品。细胞增殖率(%)=实验组的平均值/对照组的平均值×100%[1]。

[动物实验]

小鼠[2]将雌性瑞士小鼠随机分成5组(每组20只小鼠)。第1组作为对照组,第2组接受腹腔注射2.5×106埃利希腹水癌细胞,Gropu 3口服苯丁酸氮芥0.2 mg / kg体重,第4组口服左旋咪唑(5 mg / kg)体重)和第5组每天使用苯丁酸氮芥和左旋咪唑口服治疗,使用弯曲的不锈钢胃管[2]。
