AM404用途

AM404是一种内源性大麻素再摄取抑制剂,在低微摩尔范围内阻断阿难酰胺的IC50值运输[1]。AM404能够松弛用苯肾上腺素收缩的大鼠离体肝动脉,pEC50值为7.4(对应于0.04µM的EC50)。神经保护作用[2]。

[体外研究]

AM404的IC50值为4.9µM,可减少C6胶质瘤细胞增殖。AM404在用于阻止内源性大麻素anandamide的细胞聚集的浓度下,非特异性地抑制C6胶质瘤细胞增殖[2]。细胞活性测定[2]细胞系：大鼠C6胶质瘤细胞浓度：1,3,10和30µM培养时间：24,48,72和96 h结果：在暴露于10和30µM浓度的24 h和暴露于3µM浓度的48 h后,细胞增殖产生浓度依赖性降低,在72小时时,细胞增殖显著减少,尽管很小。

[体内研究]

AM404(1-5) mg/kg,i.p.)在三种模型中发挥剂量依赖性抗焦虑作用：高架+迷宫、防御性戒断和分离诱导的超声波发声。动物模型：成年雄性Sprague-Dawley大鼠(250-300只) g) [2]剂量：2.5-10 mg/kg给药：腹腔注射(i.p.)结果：导致前额叶皮质、海马和丘脑中的阿南达胺水平呈剂量依赖性增加。
