[体外研究]

Pyributicarb是一种氨基甲酸酯类除草剂,是CYP3A4基因和人类孕烷X受体(hPXR)的有效激活剂。发现Pyributicarb比典型的CYP3A4诱导剂利福平更强地增加0.1至1μM的CYP3A4报告基因活性。 hPXR-siRNA的表达明显减少了在3-1-10细胞中的Pyributicarb刺激的CYP3A4报道基因活性,并降低了HepG2细胞中内源性CYP3A4 mRNA水平[1]。 Pyributicarb通过hPXR以10nM的低浓度诱导荧光素酶转录。 Pyributicarb对hPXR的相对效力是利福平(RIF)的8.6倍[2]。

[体内研究]

Pyributicarb引起腺病毒引入hPXR的小鼠肝脏中CYP3A4衍生的报告基因活性的增强[1]。

[细胞实验]

在该实验中使用稳定表达CYP3A4报道基因(3-1-10细胞)的HepG2衍生细胞。用0.3至30μM的Pyributicarb处理细胞48小时。然后记者的活动确定[1]。

[动物实验]

使用雄性ICR小鼠(5周龄)并喂食标准啮齿动物食物。禁食18小时后,静脉内注射腺病毒[4.0×109 50%滴度培养感染剂量(TCID50)/小鼠]。感染后3天,将载体(0.5%甲基纤维素/盐水)或派瑞抑卡布(100mg / kg /天)连续2天给药。最后一次给药后20小时将动物杀死[1]。
