[体外研究]

西洛他敏是一种选择性和有效的PDE3抑制剂,PDE3A和PDE3B的IC50分别为27 nM和50 nM,具有抗血栓形成和抗内膜增生活性。西洛他敏弱抑制PDE2,PDE4,PDE5,PDE7和PDE1,IC50分别为12.5,88.8,15.2,22.0和>300μM。 Cilostamide有效抑制凝血酶诱导的血小板聚集(IC50,1.1μM)[1]。

[动物实验]

在该测定中研究血小板聚集。洗涤的血小板（200μL含有3×108个细胞/ mL的Tyrode HEPES缓冲液，pH 7.4的悬浮液）在存在或不存在不同浓度的OPC-33540，OPC-33536和37℃下在37℃温育3分钟。单独使用西洛酰胺，或与3nM PGE1组合，然后与5μL2单位/ mL凝血酶在37℃温育5分钟。使用PAM-8C聚集计测量在5分钟内透射的光强度。通过比较最大聚集率和对照值[1]来计算抑制率。
