嗪草酮制备

3,3-二甲基丁酸-2-酮的制备 以3,3-二甲基丁酮-2为原料,经氯化、水解(重排)、氧化、中和4步反应制得。



硫代卡巴肼的制备 以水合肼和二硫化碳为原料,按一定配比进行加成和脱硫化氢反应,制得硫代卡巴肼,收率85%~90%。
嗪草酮的合成 将上述合成的3,3-二甲基丁酸-2-酮与硫代卡巴肼,在催化剂存在下,进行环合反应,得4-氨基-6-特丁基-3-巯基-1,2,4-三嗪-5-(4H)-酮;此酮式与烯醇式在一定条件下存在动态平衡,加碱使酮式转化烯醇式;再与溴甲烷反应得嗪草酮。
嗪草酮的合成也可将硫代羰基肼盐酸盐溶解在二甲亚砚中,与2-特丁基氨基-3,3-二甲基丁腈反应1h,冷却至20℃,注入冰水,搅拌1h,乙醇重结晶,得3-硫基-4-氨基-5-亚氨基-6-特丁基-1,2,4-三嗪(m.p.181℃)。将其与乙醇、酸在100℃反应2h,冷却至20℃,分离结晶物,过滤,干燥,得3-硫基-4-氨基-6-特丁基-1,2,4-三嗪-5-酮(m.p.215~217℃)。最后在氢氧化钠和甲醇的混合物中,于0℃加磺甲烷,在20℃搅拌反应4h,产物经结晶、过滤、干燥得嗪草酮。

可将硫代羰基肼盐酸盐溶解在二甲亚砜中,与2-特丁基氨基-3,3-二甲基丁腈反应,冷却、搅拌、重结晶得3-硫基-4-氨基-5-亚氨基-6-特丁基1,2,4-三嗪。将其与乙醇、酸反应,分离结晶物,过滤、干燥,得3-硫基-4-氨基-6-特丁基1,2,4-三嗪-5-酮。最后在氢氧化钠和甲醇的混合物中,加碘甲烷反应,产物经结晶、过滤、干燥得嗪草酮。
