[体外研究]

在纳摩尔浓度下,内皮素-2表现出强烈且持久的血管收缩活性,特征是内皮素(EC50 = 0.52 nM)[1]。内皮素-2是巨噬细胞和THP-1单核细胞的化学引诱物,但不是新鲜分离的单核细胞。对内皮素-2的趋化反应显示出典型的钟形响应曲线。内皮素受体拮抗剂的实验表明,通过ET-RB受体介导向内皮素-2的迁移。肿瘤内皮素-2的表达可能调节巨噬细胞的行为,使活化的细胞在缺氧区域积聚[2]。

[体内研究]

两种不同的升压效应阶段通常见于内皮素-2：早期阶段紧随抑郁症反应,并在接下来的3-10分钟内占主导地位,而晚期升压效应在注射后10-20分钟发展并持续> 1小时[1]。

[动物实验]

大鼠[1]对于升压活性的测定，用氨基甲酸乙酯（1g / kg ip）麻醉重300-350g的雄性Wistar大鼠，并用阿托品（0.25mg / kg iv），普萘洛尔（1mg / kg iv）预处理。和bunazocine（1 mg / kg iv）。每3-4小时施用额外的维持剂量（初始剂量的一半）自主阻塞剂。每只动物仅给予一次内皮素-2大丸剂。从插入右股静脉的插管推注注射50μL缓冲液中的1nM内皮素-2。从套管记录血压，套管置于右颈动脉并连接到压力传感器[1]。
