DB2115 is a small-molecule transcription factor PU.1 inhibitor abrogating DNA binding by PU.1 with Kd of 1.0 nM; decrease cell growth (IC50=3.4 uM) and the clonogenic capacity of AML cells and leads to increased apoptosis, while showing little effect on normal hematopoietic cells; decreases tumor burden and increases survival in murine and human AML transplantation models.
