In Vitro 在体外实验中,Me-Indoxam对各种 sPLA2 亚型表现出不同的效力,对 hGIIA、hGIIE 和 hGV 的 IC50 均小于 100 nM;对 hGIB 和 hGX 的IC50 在 200-600 nM 之间;对 hGIID、hGIIF、hGIII 和 hGXIIA 的 IC50 大于 2 mM[1]。 ,Me-Indoxam 仅当 sPLA2s 分泌在细胞外间隙时才能够抑制其活性[1]。 Me-Indoxam (0-10 μM; 30 min) 抑制 anti-IgE 刺激的原发人肺肥大细胞 (HLMCs) 的 Leukotriene C4 (LTC4) 的产生[1]。 Me-indoxam (0.01–10 M) 不影响人肺巨噬细胞 (HLM) TNF-α 和 IL-6 的基础分泌。hGX (1 μg/mL) 与不同浓度的 Me-indoxam 预孵育 (15 min, 37°C) 后再加入 HLM,剂量依赖性地抑制 TNF-α 和 IL-6 的释放。在 TNF-α 和 IL-6 释放时,Me-indoxam 的IC50 值分别为 253±72 和 320±87 nM[2]。
