Biological Activity

[Description]:

Cyanine 3.5 (Cy3.5 NHS ester) chloride is an analog of Cy3.5 fluorophore. Cyanine 3.5 chloride is a reactive, red fluorescent dye. Cyanine 3.5 chloride is used for labeling of amino-groups in peptides, proteins, and oligonucleotides. (λex=591 nm, λem=604 nm)[1][2].

[In Vitro]

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).Labeling of Cells:1. Culture HepG2 cells in DMEM/HIGH GLUCOSE medium (containing 10% FBS, 100 U/mL penicillin G and 100 μg/mL streptomycin sulfate) at 37oC in 5% CO2 atmosphere.2. Add 1×106 HepG2 cells with logarithmic half-life in a confocal culture dish. Incubate cells at 37oC, 5% CO2, for 36 h.3. Add 200 μL of 10 μg/mL Hoechst fluorescent dye 33342. Incubate cells at 37o, 5% CO2, for 20 minutes.4. Changed the medium with 200 μL of Cy3.5-loaded FA-PEG-RH nanomicelles.5. Take cell photos with a fluorescence microscope under 350 nm and 550 nm fluorescence excitation after 1h, 4 h, 8 h, and 16 h, respectively.
