Abstract: The binding site of the antibiotic vancomycin for the peptide cell-wall analogue Ac- D-Ala-D-Ala has been studied by nuclear Overhauser effect difference spectroscopy (NOEDS). Intramolecular nuclear Overhauser effects (NOEs), observed between protons of the antibiotic in the 'H spectrum of the vancomycin/Ac-D-Ala-D-Ala complex, confirm the formation of a carboxylate binding pocket in the bound state of the antibiotic.
