| 体外研究 | 反式AUCB(t-AUCB;25-300μM;48小时)以剂量依赖的方式抑制U251和U87细胞的生长[1]。反式AUCB(200μM;48或96小时)诱导U251和U87细胞G0/G1期阻滞[1]。反式AUCB(200μM;10min-4h)可使p65的磷酸化水平在10min后升高,30min后达到高峰,持续至少2小时[1]。反式AUCB(200μM;48小时)通过激活NF-jB-p65抑制U251和U87细胞生长[1]。反式AUCB(10μM;30m in)能有效抑制人胶质母细胞瘤细胞株(U251,U87)和人肝癌细胞株(HepG2)的sEH活性[1]。细胞活力测定[1]细胞系:U251、U87细胞浓度:25、50、100、200或300μM培养时间:48小时结果:抑制U251和U87细胞生长呈剂量依赖性。细胞周期分析[1]细胞系:U251,U87细胞浓度:200μM培养时间:48或96小时结果:U251和U87细胞周期G0/G1期阻滞。Western Blot分析[1]细胞株:U251,U87细胞浓度:200μM孵育时间:10min,30min,1h,2h,4h结果:p65磷酸化水平在10min后升高,30min后达到高峰,持续至少2小时。 |
| 体内研究 | 反式AUCB(t-AUCB;p.o.;0.1,0.5,1 mg/kg)以剂量依赖的方式改善LPS诱导的低血压[2]。反式AUCB(p.o.;0.1,0.5,1 mg/kg)的t1/2值为20,30,15 min,p.o.的Cmax值为30,100,150 nmol/L,0.1,0.5,1 mg/kg[2]。反式AUCB(s.c.;1,3,10 mg/kg)的t1/2值为60,85,75 min,对于1,3,10 mg/kg的s.c.[2],Cmax值为245,2700,3600 nmol/L。反式AUCB(i.v.;0.1 mg/kg)的t1/2值为70 min和10 h,用于分布(α)和消除(β)相。反式AUCB的CL为0.7l/h•kg,Vdss为17l/kg[2]。动物模型:小鼠(雄性CFW株,7周龄,24-30g;雄性C57BL/6株,8周龄,22-25g)[2]剂量:0.1,0.5,1mg/kg给药:PO结果:LPS引起的低血压呈剂量依赖性改善。动物模型:小鼠(雄性CFW株,7周龄,24-30g;雄性C57BL/6株,8周龄,22-25g)[2]剂量:0.1,0.5,1 mg/kg(药代动力学分析)给药:p o结果:0.1,0.5,1 mg/kg的p.o.的t1/2值分别为20,30,15 min和Cmax值分别为30,100,150 nmol/L。 |
