• 体内研究: Silvestrol(1.5mg/kg,ip)不会对小鼠中异种移植的B淋巴细胞产生人IgG产生不利影响。与载体相比,Silvestrol显着延长了存活期。在任何Silvestrol处理的小鼠的脾脏中都没有检测到这种淋巴细胞浸润,这些动物在尸检时也没有表现出任何其他明显的淋巴瘤迹象[5]。
• 激酶实验: Apo-ONETM同源caspase-3/7检测试剂盒用于测量caspase-3和-7的活性。将细胞（7×10 4细胞/ mL）用Silvestrol（15nM，30nM，60nM，120nM和240nM）或依托泊苷在黑色96孔板中处理24小时。已知Etoposide在LNCaP细胞中激活胱天蛋白酶-3/7，用作该测定的阳性对照。在处理结束时，将裂解缓冲液和底物（Z-DEVD-罗丹明110）混合并加入细胞中。在通过胱天蛋白酶-3和-7活性连续切割和去除DEVD肽并在499nm激发时，罗丹明110离去基团变得强烈发荧光。发射最大值为521nm。产生的荧光产物的量与样品中存在的胱天蛋白酶-3和-7裂解活性的量成比例。样品一式三份测量。 Caspase-3和-7活性由净荧光指示[2]。
• 细胞实验: 将细胞以7×10 4个细胞/ mL的密度接种在100-mm培养皿中，并用30nM或120nM浓度的Silvestrol处理24小时。将细胞用胰蛋白酶消化，用PBS洗涤，并用PBS中的1％（w / v）多聚甲醛在冰上固定30分钟。离心后，将细胞在-20℃下悬浮于70％（v / v）乙醇中直至使用。从反应管中除去所有乙醇，用PBS洗涤细胞两次。处理细胞用于在22℃-24℃下用荧光素标记的脱氧尿苷三磷酸核苷酸标记过夜，洗涤，并在室温下与碘化丙锭/ RNase A溶液在黑暗中孵育30分钟。然后通过流式细胞术分析标记的细胞[2]。
• 动物实验: 小鼠[5]通过用抗去唾液酸（GM1）预处理（加上每周再治疗），将外周血单核细胞（PBMC）腹膜内（IP）注射到耗尽小鼠自然杀伤（NK）细胞的SCID小鼠中。通过hu-IgG ELISA确认植入。在植入后2周开始用载体（30％羟丙基-β-环糊精）或Silvestrol（每48小时IP 1.5mg / kg）处理[5]。
