体内研究 含有二硬脂酰磷脂酰胆碱(DSPC)的脂质纳米颗粒(LNP)和可离子化的脂肪-脂质如二亚油基甲基-4-二甲基氨基丁酸酯(DLin-MC3-DMA)是体内有效的siRNA递送载体。 LNP-siRNA系统优化以在肝细胞中实现最大的基因沉默效力,然后在小鼠中静脉内施用含有DLin-MC3-DMA(MC3),DSPC,胆固醇和聚乙二醇(PEG)-脂质,摩尔比为50/10/38.5/1.5。 DLin-MC3-DMA具有优化的pKa值,可显着提高效力[1]。
