[体外研究]

自由基清除活性随2-吡咯烷酮用量的增加而增加。发现2-吡咯烷酮的细胞毒性具有剂量和时间依赖性,并在所有选定的癌细胞系中通过显微镜观察其作用。观察到2-吡咯烷酮对HeLa的细胞毒性浓度分别为2.5 mg/mL,对PC-3细胞的细胞毒性浓度为3 mg/mL,对HeLa和PC-3细胞的细胞毒性浓度分别为1.5 mg/mL和2 mg/mL,对PC-3细胞的细胞毒性浓度分别为48 h。随着纯化2-吡咯烷酮浓度的增加,细胞活力降低。用2-吡咯烷酮(其IC50浓度分别为1.5 mg/mL和2 mg/mL,用于HeLa和PC-3细胞)处理24小时后,观察细胞的形态学变化。研究表明,2-吡咯烷酮通过细胞周期阻滞的G0/G1期诱导HeLa和PC-3细胞凋亡[1]。

[细胞实验]

使用MTT法评估抑制浓度(IC50)。将癌细胞(1×104细胞/孔)在96孔培养皿中培养48小时,形成75%的浓度。用浓度为(0.5至5 mg/mL)2-吡咯烷酮的新鲜培养基替换培养基,并将细胞进一步培养24小时和48小时。移除培养基,向每个孔中添加100 mL MTT溶液,并在37°C下培养4小时。去除上清液后,向每个孔中添加50 mL二甲基亚砜,并孵育10分钟以溶解福尔马赞晶体。在ELISA多孔板读取器中在620nm处测量光密度。OD值用于计算生存能力的百分比[1]。
