[体内研究]

硫柳汞(THIM)给药以剂量依赖性方式增加中脑导水管周围灰质(PAG)中的μ-阿片受体(MOR)密度。高剂量硫柳汞治疗显著增加中脑导水管周围灰质背内侧区(DMPAG)和中脑导水管周围灰质外侧区(LPAG)的MOR密度。在剂量为3000μg Hg/kg时,硫柳汞还会增加尾壳核(CPU)中的MOR密度。相反,两种较高剂量的硫柳汞均能降低齿状回(DG)的MOR密度[1]。硫柳汞给药(出生后第7、9、11、15天注射4次,即240μg Hg/kg)可引起氨基酸溢出的持续变化:谷氨酸和天冬氨酸增加,同时甘氨酸和丙氨酸减少;注射后10至14周测量。微透析时,以12.5μg Hg/kg的剂量四次注射硫柳汞不会改变谷氨酸和天冬氨酸的浓度。在灌注液中将硫柳汞应用于前额叶皮质(PFC)会引起谷氨酸溢出的快速增加。神经甾体、硫酸脱氢表雄酮(DHEAS;80mg/kg;i、 防止硫柳汞对谷氨酸和天冬氨酸的影响;单用类固醇对这些氨基酸没有影响。在灌注液中同时应用硫酸脱氢表雄酮(DHEAS)和硫柳汞也可阻断硫柳汞对谷氨酸的急性作用[2]。

[动物实验]

本研究采用雄性大鼠。在出生后第20周,动物被戊巴比妥过量安乐死,然后斩首。用两种更高剂量的硫柳汞(THIM)(1440和3000μg Hg/kg)治疗的大鼠来自原始幼仔,用于不同的行为和病理测试,但在实验上是幼稚的。根据相同的程序,在出生后第8周处死一组单独的实验性天真大鼠,用两种较低剂量的硫柳汞(12和240μg Hg/kg)(每种药物治疗4窝)治疗。每个实验组由5只动物组成,最多2只来自同一窝动物[1]。
