[体外研究]

Mag-Indo-1四钾盐可用于测定细胞质溶液中的游离Mg2+浓度[1]。指南(以下是我们推荐的方案。该方案仅提供指南,应根据您的具体需要进行修改)[1]。1.通过混合两种储备溶液的不同比例来制备含有不同浓度Mg2+的校准溶液：一种含有(mM)150 KCl、0.1 EGTA和5 Hepes(KOH的pH值为7.2),另一种含有100 MgCl2、0.1 EGTA和5 Hepe(KOH为pH值为7.2)。2.使用含有1μM Mag-Indo-1四钾盐的校准溶液构建校准曲线。3.所含“Mg2+”校准溶液(mM)：130 KCl、4 EDTA和5 Hepes(pH 7.2,KOH)。4.荧光比(R)的背景校正值与Mg2+浓度之间的关系符合以下理论方程。[Mg]=K(R-Rmin)/(Rmax-R),其中[Mg]是游离Mg2+离子的浓度,Rmin是0[Mg2+]时的R值,Rmax是Mg2+饱和时的R数值。曲线拟合得出Rmin=0.053,Rmax=1.57,K=5.1 mM。注：具体曲线可参考参考。
