[体外研究]

Exendin-3与豚鼠胰腺腺泡中的至少两种受体相互作用;在高浓度(> 100nM)下,肽与VIP受体相互作用,从而引起cAMP的大量增加和刺激淀粉酶释放;在较低浓度(0.1-3nM)下,肽与推定的毒蜥外泌肽受体相互作用,从而导致未确定功能的cAMP的较小增加[1]。

[激酶实验]

用VIP,促胰液素或毒蜥外泌肽-3观察到增加浓度的两种VIP受体拮抗剂对淀粉酶释放增加的影响。将雄性Hartley豚鼠Acini与VIP(1nM),促胰液素(1μM)或Exendin-3(1μM)在37℃温育30分钟,单独或与指定浓度的[AcTyr,D-Phe]组合] GRF 1-29酰胺(实心符号)或[4-Cl-D-Phe,Leu] VIP(空心符号)[1]。
