[体外研究]

与细胞接触时,TMA-DPH瞬间掺入质膜中。 TMA-DPH浓缩在溶酶体和高度酸性的晚期内体中[1]。 TMA-DPH荧光寿命保持恒定,6.2±0.2 ns,低于相同的2μM临界浓度,但随着浓度的增加而显着降低。然而,这种下降速度减慢到5μM以上。 TMA-DPH荧光各向异性显示出与荧光寿命类似的演变。它首先是常数,0.283±0.003,再次直到2μM,然后在5μM时迅速下降至0.270±0.003,继续下降,但浓度较高时速率较低[2]。

[细胞实验]

使2mL DM10F中的L929细胞粘附在显微镜载玻片上。为了观察质膜标记，将细胞在室温下用TMA-DPH 2×10-6M的PBS溶液或DM10F从4×10-3M的二甲基甲酰胺储备液中培养短时间（10秒）。然后将未洗涤的载玻片转移到显微镜中并观察。对于内化膜的标记，将细胞在37℃的载玻片中，在DM10F中用TMA-DPH 2×10-6M孵育所需的时间，然后通过在PBS中轻轻摇动载玻片几秒钟来洗涤。 [1]。
