[体外研究]

淀粉样蛋白β-肽(1-42)人是42-氨基酸肽,其在阿尔茨海默病的发病机理中起关键作用。淀粉样β-肽(1-42)人(1至10μM)在沐浴液中的应用不会改变延迟整流器K +电流和漏电流,但会增强Са2+电流的失活并阻断Са2+依赖的К+电流[1]。在2.5μM浓度下,淀粉样蛋白β-肽(1-42)人将SH-SY5Y细胞的存活率降低至65％。结果表明,淀粉样蛋白β肽(1-42)在孵育30分钟后和8小时后定位于SH-SY5Y细胞的细胞质和细胞核中。在后者中,在细胞质和细胞核中观察到大量淀粉样蛋白β-肽(1-42)。在淀粉样蛋白β-肽(1-42)人治疗中也检测到APP mRNA水平增加[2]。

[细胞实验]

用生物素化的淀粉样蛋白β-肽（1-42）人肽1μM处理SH-SY5Y细胞。将细胞固定并用含有0.5％Triton X-100的3.3％甲醛透化，然后用含有镁和钙的PBS中的125mM甘氨酸透化。用5％胎牛血清血清封闭细胞，然后用第一抗体封闭。用单克隆抗体AB或用Avidin Fluor488检测生物素 - 淀粉样蛋白β-肽（1-42）人肽。细胞核用DAPI染色。使用LSM 510 meta共聚焦显微镜[2]获得图像。
