[体外研究]

在SDS-PAGE后检测丹酰基尸胺荧光显示在丹酰基尸胺和转谷氨酰胺酶存在下肌动蛋白-胸腺素β4交联的量非常低,同时存在大量丹酰基尸胺标记的胸腺素β4和肌动蛋白。在丹酰基尸胺存在的情况下,交联量进一步降低,竞争G-肌动蛋白或胸腺素β4(100μM)的Lys残基。在没有胸腺素β4的情况下,丹磺酰尸胺标记的肌动蛋白聚合成F-肌动蛋白,如通过天然PAGE [1]后的荧光检测所揭示的。

[激酶实验]

为了确定转谷氨酰胺化的时间过程，将胸腺素β4（120μM）与丹酰基尸胺（5mM）在由10mM Tris-HCl，pH 7.4,15mM CaCl 2,3mM DTT组成的70μL缓冲液中温育。通过加入0.1U转谷氨酰胺酶开始反应。在加入酶（t = 0）后立即在指定时间，从混合物中取出10μL，在490μL0.1％TFA中稀释以终止反应并通过HPLC分析[1]。

[细胞实验]

将MCF7细胞（2.4×104）接种到35mm平板中。温育24小时后，在存在或不存在3-甲基腺嘌呤（3-MA）的情况下，以不断增加的浓度添加CuO NPs不同的时间段。然后将细胞与50mM丹酰基尸胺（MDC）在37℃温育15分钟，并用1×PBS洗涤三次，间隔5分钟。最后，在荧光显微镜下观察细胞[2]。
