Palmitoyl Tripeptide-1 (INCI: Palmitoyl Tripeptide-1), also known as Pal-GHK or Biopeptide CL, is one of the most extensively researched and clinically validated signal peptides in cosmetic science. Its sequence is Pal-Gly-His-Lys-OH (N-(1-oxohexadecyl)glycyl-L-histidyl-L-lysine), with a molecular weight of approximately 578.79 Da and molecular formula C30H54N6O5. Originally developed by Sederma SAS (now part of Croda International), Pal-GHK is a synthetic lipopeptide derived from the naturally occurring GHK tripeptide, a fragment of the alpha-2(I) chain of type I collagen first identified by Pickart in 1973. The palmitic acid (C16) chain was introduced to dramatically increase lipophilicity -- by 100 to 1000-fold -- enabling efficient penetration through the lipid-rich stratum corneum to the dermis where fibroblasts reside. Pal-GHK functions as a matrikine signal peptide that mimics collagen breakdown fragments. When fibroblasts detect the GHK sequence, they interpret it as evidence of collagen degradation and respond by upregulating synthesis of new ECM components through the TGF-beta signaling pathway. In vitro studies show that at 0.5 muM, Pal-GHK increases collagen I synthesis by up to 100% in human dermal fibroblasts. Clinical studies demonstrate progressive improvement: 23% wrinkle depth reduction at 4 weeks, 44.9% at 8 weeks, and up to 68% at 6 months. Skin elasticity improved by 18.81%, hydration by 28.12%, and collagen production by 54.99% after 12 weeks. |
