[体外研究]

Bergamottin是CYP1A1的竞争性抑制剂。 Bergamottin抑制CYP1A1,CYP1A2,CYP2B1和CYP2B2,IC50分别为0.192±0.029μM,5.077±0.31μM,9.495±0.979μM,4.535±0.092μM[1]。 Bergamottin对A549细胞具有有效的抗增殖作用。 Bergamottin显示出对这些细胞的浓度依赖性和时间依赖性生长抑制作用。 Bergamottin还通过减少癌症集落形成细胞的数量来抑制A549癌细胞的克隆形成活性。克隆形成的减少也遵循浓度依赖于Bergamottin [2]。

[体内研究]

使用雌性BALB/c裸鼠(总共20只小鼠)测定了Bergamottin在体内条件下的抗癌功效。通过注射非小细胞肺癌A549细胞(1×10 6个细胞/小鼠)在小鼠中诱导肿瘤。在肿瘤形成后,处死小鼠并除去肿瘤并计算它们的重量和体积。结果显示25,50和100mg/kg佛手柑注射液将PBS处理组(对照)中的肿瘤重量从1.61g减少至1.21,0.42和0.15g。与载体组相比,最高浓度的Bergamottin组(100mg/kg体重)中裸鼠的肿瘤重量显着降低(P <0.05)。同样地,25,50和100mg/kg的Bergamottin注射剂将PBS处理组(对照)中的肿瘤体积从2.2cm 3减少至1.71,1.1和0.51cm 3。肿瘤异种移植物体积的定期测量表明,与载体组相比,最高浓度的Bergamottin组(100mg/kg体重)中裸鼠的肿瘤体积显着减少(P <0.05)[2]。

[激酶实验]

CYP1A1超级体和不同浓度的7-乙氧基脱嘌呤（ER）用于测定CYP1A1酶促动力学。找到典型的Michaelis-Menten曲线。最终的反应混合物含有：1pmol CYP1A1,0.5mM NADPH，不同的ER浓度和0,4,8和16nM的Bergamottin（BG）。加入NADPH开始反应。通过对具有竞争型抑制的Michaelis-Menten方程拟合的实验数据的非线性回归分析获得动力学常数。通过使用Lineweaver-Burk，Dixon和Dixon曲线斜率的重绘图也显示了动力学分析[1]。

[细胞实验]

通过MTT测定法测量Bergamottin对细胞增殖的抑制。简而言之，将人肺腺癌A549细胞接种在96孔培养板（1×10 5细胞/孔）中。温育24小时后，用Bergamottin（0,5,10,25,50,75和100μM）处理细胞24和48小时，然后向每个孔中加入MTT溶液（10mg / mL）。孵育4小时后，将甲crystals沉淀物溶解在100μLDMSO中，然后在自动化微量化读数器中在570nm处测量吸光度。计算细胞存活率。细胞毒性表示为抑制细胞生长50％所需的Bergamottin浓度（IC50值）[2]。

[动物实验]

小鼠[2]雌性BALB / c裸鼠（6周龄）（总共20只）在无病原体的环境中随意保持水和食物，在动物中12小时光照和12小时黑暗循环护理设施。将人非小细胞肺癌A549细胞（2×10 6个细胞/小鼠）注射到裸鼠的右腋窝（5只小鼠/组）中以在小鼠中产生肿瘤。在肿瘤发展之后，将小鼠分成4组并用腹膜内注射的Bergamottin处理。研究中的对照组用等量的PBS处理，而其他三组用25,50和100mg / kg的Bergamottin处理。然后，在18天后处死小鼠，评估每只小鼠的肿瘤重量和体积。使用游标卡尺测量肿瘤长度和宽度，并计算肿瘤体积（TV）[2]。
