[体外研究]

在MC65细胞中,一种细胞AD模型,薯di皂苷元在1μM时表现出较弱的保护能力,可能代表我们抗AD病理的二价化合物中作为甾体部分的良好候选[1]。薯gen皂苷元以剂量依赖的方式抑制C3A细胞中STAT3的组成型活化,最大抑制发生在100μM左右[2]。

[体内研究]

薯gen皂苷元治疗显着降低HF饮食喂养小鼠的禁食和补充血糖水平。与HF饮食喂养的小鼠相比,薯gen皂苷元治疗显着降低了IPGTT试验中的血糖水平[3]。

薯蓣皂苷元制备

1.皂素广泛存在于植物界，单子叶植物和双子叶植物中均有分布，尤以薯芋科、百合科、石竹科、五加科、豆科、七叶树科、远志科、桔梗科、玄参科等植物中含量较高。主要采用的生产原料为薯芋科植物穿龙薯芋和盾叶薯芋(黄姜)。

将薯芋干品浸泡，粉碎成拇指盖大小颗粒，再上磨浆机带水磨成浆状，过筛分出纤维渣，过滤液静置。此处磨浆一定要细，要将纤维和淀粉分开，不能使纤维中含有淀粉物，否则皂甙将损失而影响提取得率。静置的过滤液分出上层清液，然后浓缩上清液至浆状，再用乙醇提取，乙醚沉淀，得到可用于治疗冠心病的穿龙冠心宁。分去上清液的下层淀粉液加入淀粉酶，使淀粉转化成葡萄糖，过滤，滤渣用盐酸水解，水洗，干燥可制得薯芋皂素。
