MPP+ iodide用途

MPP+ iodide 是神经毒素 MPTP 的一种有毒代谢物,通过选择性破坏黑质多巴胺能神经元,在动物模型中引起帕金森病症状。MPP+ iodide 被多巴胺转运体吸收进入多巴胺能神经元,通过影响呼吸链复合体 I 对线粒体发挥神经毒性作用。MPP+ iodide 也是 5-羟色胺转运体 (SERT) 的高亲和力底物。

[体外研究]

MPP+(1-3 mM；24小时)显著降低细胞活力[1]。MPP+还抑制线粒体制剂中烟酰胺腺苷二核苷酸(NADH)连接呼吸的活性,并损害有氧糖酵解,因此目前认为NADH连接细胞呼吸的抑制可能构成MPP+神经毒性的最终分子机制[3]。细胞活力测定[1]细胞系：SH-SY5Y细胞浓度：1,2,3 mM孵育时间：24小时结果：用MPP+处理SH-SY5Y细胞,模拟PD时多巴胺能神经元丢失的过程；细胞活力呈剂量依赖性(1,2,3 mM孵育24小时)和时间依赖性(1 mM)。
