[体外研究]

AEBSF通过直接抑制神经和非神经五种不同人类细胞系中的β-分泌酶来抑制Aβ的组成型生成[1]。 AEBSF作为丝氨酸蛋白酶抑制剂,抑制人巨噬细胞对白血病细胞的溶解,而不抑制TNF-α和IL-1β的巨噬细胞分泌[2]。 AEBSF还通过改变蛋白质分泌模式来干扰胚泡在子宫内膜细胞上的生长并抑制HeLa细胞对HUVEC的粘附[4]。

[体内研究]

AEBSF(每天76.8 mg/kg,ip)导致已经给予致死型弓形虫感染的小鼠的存活延长[3]。 AEBSF还可减少蟑螂过敏原诱导的小鼠模型中的气道反应和潜在炎症[4]。

[细胞实验]

将悬浮在含有10％FCS的RPMI-1640培养基中的HeLa细胞接种到96孔微孔板（5×103细胞/200μL/孔）的每个孔中。在37℃温育24小时后，用不同剂量的AEBSF（0,25,50,100μg/ mL）处理细胞48小时。然后，向每个孔中加入20μL新鲜的3-（4,5） - 二甲基噻唑（-z-y1）-3,5-二苯基四唑鎓（MTT）试剂（5μg/μL），并在37℃下培养细胞。在5％的二氧化碳中再持续4小时。小心丢弃培养基，并加入150μLDMSO。在酶标仪上以540和620nm的双波长读取吸光度。

[动物实验]

根据给予的治疗将注射2.5×103寄生虫的小鼠随机分配至一个治疗组：不含药物（对照组），单独载体（载体对照组），不同剂量的单独乙胺嘧啶，不同剂量的单独LY311727，AEBSF单独使用不同剂量或AEBSF 76.8 mg / kg加乙胺嘧啶10 mg / kg。每个治疗组由10只动物组成。在寄生虫接种后24小时开始治疗，并持续连续七天。每天监测小鼠存活并在活小鼠中继续存活直至感染后15天。所有实验均进行三次，所示数据代表累积结果。
