[体外研究]

当分析代表DNA损伤程度的尾部力矩和尾部强度时,仅测试的两种最高浓度的赤藓红B(50.0和70.0μg/ mL)与载体对照组显着不同(p <0.05)。结果显示,与载体对照组相比,7种Erythrosin B浓度中的6种(0.2至70.0μg/ mL)的微核(MNi)频率增加[1]。

[细胞实验]

进行碱性单细胞凝胶电泳测定(彗星测定)。简而言之,将2×105个HepG2细胞接种在24孔板中24小时。然后用Erythrosin B以0.1,0.2,2.0,10.0,25.0,50.0或70.0μg/ mL(终浓度)处理细胞4小时;还进行载体对照(0.7%DMSO)和阳性对照(多柔比星0.3μg/ mL)处理。将HepG2细胞悬浮液与37℃低熔点琼脂糖混合,并转移至正常熔点琼脂糖涂覆的载玻片上。每次处理分析100个随机选择的类核,并进行总共三次独立实验[1]。

赤藓红B制备

1.赤藓红的制备

将间苯二酚加热熔融,加入邻苯二甲酸酐搅匀熔化,加热至185℃反应半小时,生成深黄色的间苯二酚酞,此时呈液体状态。在搅拌下加入粉末状无水氯化锌,搅拌至完全溶解,并逐步升温至210~215℃,加热至反应物完全固化,再冷却、粉碎得粗融物;将此粗融物与稀盐酸加热共沸,浸出氯化锌和残留的间苯二酚,过滤、水洗、干燥后,再用乙醇提取未反应的间苯二酚酞,提取过的产物经干燥即得荧光素。

将荧光素溶解于氢氧化钠溶液中,加碘进行反应,反应物中加入盐酸,析出结晶,然后将其转化为钠盐溶液,浓缩即得产品。 赤藓红铝色淀的制备。

由氯化铝、硫酸铝等的铝盐与碳酸钠等的碱类制取氢氧化铝,然后加于赤藓红水溶液中,沉淀而得产品。

2.由苯甲醛-4-磺酸与二乙基间氨基苯酚缩合生成的产物,经浓硫酸脱水,用重铬酸钾、高锰酸盐或氯化铁氧化,制成钠盐。

3.由间苯二酚、邻苯二甲酸酐及无水氯化锌加热熔融而得的粗制荧光素,经精制、碘化后,用氯化钠盐析,再经精制而得。

4.以荧光黄为原料与碘反应后加氢氧化钠成钠盐,经精制、过滤得成品。
