[体外研究]

儿茶素对环氧合酶-1(COX-1)的抑制活性在70μg/ mL时> 95%,IC50为1.4μM[1]。在用儿茶素处理24小时后观察到剂量依赖性的颜色减少,并且在最高浓度的儿茶素测试(160μg/ mL)时54.76%的细胞死亡,而儿茶素的IC50达到127.62μg/ mL儿茶素。当用儿茶素处理MCF-7细胞时,观察到细胞凋亡诱导的剂量和时间依赖性增加。当与对照细胞在24小时比较时,分别用150μg/ mL和300μg/ mL儿茶素处理的细胞的40.7和41.16%经历凋亡。用150μg/ mL儿茶素处理24小时后,MCF-7细胞中Caspase-3,-8和-9及p53的表达水平分别比5.81,1.42,3.29和2.68倍增加。在未经处理的对照细胞中[2]。

[体内研究]

用最低测试剂量(即50mg/kg)的儿茶素治疗的动物在选择试验中花费相对更多的时间来探索新物体,然而,差异在统计学上不显着。儿茶素以剂量依赖的方式防止时间诱发的情景记忆缺陷,最有效的是200mg/kg,po。与单独DOX治疗组相比,儿茶素治疗可预防MPO水平升高(海马和额叶皮质分别为21.98±9.44和36.76±4.39%)[3]。

[细胞实验]

进行细胞活力测定以评估不同浓度的儿茶素对MCF-7细胞的毒性。简而言之,将MCF-7细胞(2×10 4个细胞/孔)接种在96孔板中,并用0μg/ mL儿茶素和160μg/ mL儿茶素处理24小时。然后,将40μLCellTiter Blue溶液直接加入孔中并在37℃下孵育6小时。使用微孔板荧光读数器以560nm / 590nm(激发/发射)滤光器组记录荧光,并计算IC 50。在三个独立实验中针对每种浓度的儿茶素运行四联体样品[2]。

[动物实验]

大鼠[3] 12周龄,体重200-230g的健康雄性大鼠用于本研究。对于一种载体和三组儿茶素(三个剂量),将大鼠分成四个实验组(每组n = 9)。儿茶素的剂量以50,100,200mg / kg在0.25%w / v羧甲基纤维素钠(CMC)中制备,并在实验试验之前和期间口服给药7天。在本研究中评估了情景记忆,对过去经历的有意识记忆[3]。
