MCL0129 is a selective, potent melanocortin-4 receptor (MC4R) antagonist with Ki of 7.9 nM, without affinity for MC1 and MC3; attenuates the alpha-MSH-increased cAMP formation in COS-1 cells expressing the MC4, does not affect basal cAMP levels; exhibits anxiolytic- and antidepressant-like activities in various rodent models.
